U1 RNP antibodies (also called anti-U1RNP) are a type of autoantibody that mistakenly targets the body’s own U1 ribonucleoprotein (U1 RNP) complex, a critical component of the spliceosome involved in processing messenger RNA. This test measures the level of these antibodies in the blood, usually by indirect immunofluorescence or ELISA, and is reported as a titer or concentration.
The presence of anti-U1RNP antibodies is a hallmark of mixed connective tissue disease (MCTD, or Sharp syndrome), a condition with overlapping features of lupus, scleroderma, and polymyositis. However, they can also be found in some patients with systemic lupus erythematosus (SLE) and other autoimmune disorders. Testing is used to aid in the diagnosis and differentiation of connective tissue diseases, especially when a patient presents with symptoms such as Raynaud’s phenomenon, joint pain, or skin changes.
